若转膜指示条带(如预染Marker)缺失或异常,请检查转膜缓冲液、电极连接及转膜顺序;若Marker正常,则需排查样本中目的蛋白是否存在、是否降解,或验证抗体特异性及稀释比例。
建议降低蛋白上样量、抗体使用浓度、ECL发光液用量或缩短曝光时间,以获取清晰适中的信号强度。
常见原因为封闭不充分、一抗浓度过高、洗膜不彻底或抗体与封闭剂存在交叉反应,建议系统优化封闭与洗涤步骤,并验证抗体特异性。
可适当延长发光液孵育时间(如2–3分钟);若为低丰度蛋白检测,建议选用更高灵敏度发光液(CW0049P)。
建议将发光液分次、少量滴加于膜上,避免一次性大面积覆盖导致过快消耗;若信号衰减明显,可补加新鲜发光液后再次成像。
若背景干净且信号持续时间正常,多为抗体浓度过高引起,需进一步稀释一抗;若背景脏、信号持续时间短,则可能与HRP过量有关,建议稀释HRP标记二抗。
此为HRP酶过量导致底物局部快速耗尽所致,建议将HRP标记抗体稀释至少10倍以上,并优化抗体孵育条件。
